百年前，玉须仙师封印八州异兽，西南神木化为枯木（化作灵源）。大赵王朝四十年，往生邪教顾念彤欲破仙师封印，炼祭神木化作树妖，荼毒生灵。为此，先帝特诏镇魔司隐监唐凌风等人，挥师西南平定妖邪。而降服树妖需要公输家所创机关巨兽，驱动巨兽的办法则是《鲁班全书》中记载的“灵源秘术”，这也因此牵连出十几年前唐家与公输家、镇魔司的血仇恩怨，数十年前，公输家为求《鲁班全书》屠尽唐家满门，侥幸逃生年仅八岁的唐凌风被镇魔司统领张谦带回镇魔司抚养，唐凌峰在此结识了陆隐、陆瞳兄妹，三人亲若兄妹，并一同通过严格考核，成为隐监成员，而随着唐凌风身世揭开，唐凌风与陆隐、陆瞳兄妹关系发生变化，三人也悄然落入公输家的阴谋中。。山田日後承認在開拍此片之前，已心諳渥美清的肺病步入末期，決定此章為最後一章，並安排由淺丘瑠璃子演的莉莉送寅次郎踏上人生的最後一程。分別15年後，兩人於如詩如畫的奄美大島重逢，感情依舊，鬥氣繼續，沒完沒了似的。正如山田所想，雖然系列必須完結，但始終不想為寅次郎的故事／人生劃上句號。結局如常開放，代表寅一直繼續他的旅程，日本人依然有夢。 公映後翌年，渥美清病逝，享年68歲，本片亦成為他的遺作。 Yamada knew before production began that Atsumi's health has deteriorated to such an extent that Tora-san to the Rescue would be the last film in the series.
